要旨:
Oxidants, which are endogenously generated within cellular and tissue environments, have been implicated as causative factors in the pathogenesis of numerous major diseases. While this paradigm remains widely accepted and oxidants undoubtedly contribute to disease etiology, it is now recognized that they also function as critical regulatory molecules essential for maintaining homeostasis in both physiological and pathological contexts. These reactive species can be detected and transduced into specific biological responses through their interactions with select cysteine thiol residues on proteins. Such interactions can induce oxidative post-translational modifications that, in certain cases, modulate protein function to facilitate cellular adaptation and homeostasis. By identifying thiol-based redox sensor proteins that undergo oxidant-mediated structural and functional alterations, we can gain a deeper understanding of the biological significance of these modifications in cardiovascular health and disease. In this presentation, I will discuss our investigations into newly identified thiol redox sensors and our efforts to develop electrophilic therapeutics targeting these proteins to address unmet clinical needs in cardiovascular disease.
